AGAMOUS a single copy genewith high potential for cycad phylogenetics.

Phylogenetic relationships within cycads have been mostly based on the Internal Transcribed Spacers of nuclear ribosomal DNA and on several regions of the chloroplast genome. There has been a paucity of single copy nuclear genes that can be useful for phylogenetic reconstructions within the Cycadales. As part of our ongoing research on molecular systematics of cycads we have developed a protocol to amplify and sequence the MADS-Box AGAMOUS gene across all the genera of the order. The aligned sequences were over 1800 bp in length. A gene tree that included representatives of all the genera of the Cycadales provided strong phylogenetic support for most of the recovered nodes.The resulting phylogeny provided insights concerning the controversia lplacement of Bowenia and Stangeria. In addition, it showed Dioon as sister to the rest of the Zamiaceae. A pilot study with a small data set of species of Zamia demonstrated the phylogenetic utility of AGAMOUS at the infrageneric level and suggested that this gene can be useful in reconstructing phylogenetic relationship of other genera.
